The connectivity stays the same while the 3-D arrangement changes. A chiral pair can be non-superimposable mirror images with different biological interactions.

The stage uses a tetrahedral centre as a spatial cue. Assigning R/S in a real molecule requires priority rules and a defined viewing orientation; rotation alone is not a full assignment procedure.
